Charles Cecil John Manners was born on 16 May 1815, the son of John Henry Manners, 5th Duke of Rutland and Lady Elizabeth née Howard. Styled Marquess of Granby before 1857, at his father’s death he gained the title 6th Duke of Rutland.

He never married and died on 4 March 1888 at the age of 72, the title passing to his younger brother.

The St James's Gazette (5 March 1888) announced 'that the Duke of Rutland died at Belvoir Castle yesterday afternoon at four o'clock. [...] Lord and Lady John Manners had been staying at Belvoir Castle since Tuesday last, owing to the continued illness of his Grace, which prevented his lordship attending the Cabinet Council held on Saturday. The late Duke of Rutland, K.B., (Sir Charles Cecil John Manners), Marquess of Granby, Earl of Rutland, and Baron Manners, was born in 1815, and succeeded to the Dukedom on the death of his father in 1857. He represented Stamford in the House of Commons from 1837 to 1852, and from that time until his succession to the peerage he sat for North Leicestershire. He was a Conservative and ardent Protectionist. He was Lord-Lieutenant and Custos Rotulorum of the county of Leicester, and honorary colonel of the 2nd battalion of the Leicester Regiment. His Grace was never married. Lord John Manners, brother of the late Duke, succeeds to the title. He was born in 1818, entered Parliament in 1841, and since 1852 has held various offices in Conservative Administrations, being at present Chancellor of the Duchy of Lancaster. By his succession to the peerage a vacancy is created in the representation of the Melton Division of Leicestershire.'
